NAD+ — Nicotinamide Adenine Dinucleotide

NAD+ (Nicotinamide Adenine Dinucleotide) is a critical coenzyme found in every living cell. This dinucleotide consists of two nucleotides joined through their phosphate groups — one containing an adenine nucleobase and the other containing nicotinamide. With a molecular weight of 663.43 Da and the molecular formula C₂₁H₂₇N₇O₁₄P₂, NAD+ serves as a fundamental electron carrier in redox reactions central to cellular metabolism.

NAD+ has become one of the most intensively studied molecules in modern biochemistry. Published research has characterized its role as a substrate for sirtuins (SIRT1-SIRT7), poly(ADP-ribose) polymerases (PARPs), and CD38/CD157 ectoenzymes. These NAD+-consuming enzymes are the focus of extensive in-vitro investigations into cellular signaling, DNA maintenance pathways, and metabolic regulation in preclinical research models.

AXOM NAD+ is manufactured under strict quality protocols and verified to ≥99% purity via HPLC analysis. Each lot is accompanied by a Certificate of Analysis documenting identity, purity, and quality metrics. Available in 500mg lyophilized format. For research and laboratory use only.

Essential Coenzyme

Critical cofactor in over 500 enzymatic reactions. Serves as the primary electron carrier in redox reactions fundamental to cellular energy metabolism research.

Sirtuin Substrate

Required substrate for sirtuin (SIRT1-7) enzymatic activity, enabling in-vitro studies of NAD+-dependent deacetylase and ADP-ribosyltransferase pathways.

Research-Grade Purity

≥99% HPLC-verified purity with full COA documentation. Every lot independently tested by third-party analytical laboratories.

NAD+ metabolism and the control of energy homeostasis: a balancing act between mitochondria and the nucleus
Cantó C, Menzies KJ, Auwerx J • Cell Metabolism, 2015 • Review

Comprehensive review examining NAD+ as a central metabolic regulator. Characterizes the coenzyme's role in mitochondrial function, nuclear signaling, and the interplay between NAD+ biosynthesis and consumption pathways across multiple in-vitro and preclinical research models.

Declining NAD+ induces a pseudohypoxic state disrupting nuclear-mitochondrial communication during aging
Gomes AP, et al. • Cell, 2013

Landmark study characterizing the relationship between NAD+ levels and nuclear-mitochondrial communication in cell culture models. Demonstrated that NAD+ modulates HIF-1α signaling through SIRT1-dependent mechanisms in controlled laboratory conditions.

NAD+ and sirtuins in aging and disease
Imai S, Guarente L • Trends in Cell Biology, 2014

Review of NAD+-dependent sirtuin biology, examining the biochemical mechanisms by which NAD+ availability regulates sirtuin enzymatic activity. Discusses in-vitro assay methodologies for measuring NAD+ flux and sirtuin function in research applications.

Reconstitution Protocol

NAD+ is supplied as a lyophilized powder and should be reconstituted with sterile water or appropriate buffer solution for research applications.

  • Allow the vial to reach room temperature before reconstitution
  • Add sterile water or PBS buffer slowly along the vial wall
  • Gently swirl — do not shake or vortex
  • Allow the solution to stand until fully dissolved
  • Solution should appear clear and colorless

Storage Conditions

  • Lyophilized (unopened): Store at -20°C for long-term stability. Stable at 2–8°C for up to 90 days.
  • Reconstituted: Store at 2–8°C (refrigerated). Use within 14 days of reconstitution.
  • Protect from light, moisture, and repeated freeze-thaw cycles.
  • NAD+ is hygroscopic — keep tightly sealed when not in use.

Shipping

All orders ship within the continental United States via USPS or UPS. Products are shipped at ambient temperature — lyophilized compounds are stable during transit. Free shipping on orders over $250.
